Abstract

The genome of Chrysemys picta belli has been recently sequenced. The main objective of our project is to find out all the known homologous selenoproteins of the genome. Selenoproteins are a family of proteins that are distinguished for containing selenocisteine, the 21st aminoacid. These proteins become relevant in many essential cellular processes such as oxidative stress defense or immunological response activation. Selenocisteine is codified by a TGA codon, wich in normal conditions is an STOP codon. Several structures such as SECIs and other enzimes are required to allow Selenoproteins translate. The difficulties of this specific type of traduccion has been a strong handicap in the annotation process of selenoproteins. We have designed a serial group of programes that integrates several tools such as BLAST, Exonerate, GeneWise, T-Coffee, SECISearch and BLAST against non-redundant NCBI database in order to indentify and anotate all the existing proteins in Crysemys picta belli genome. The results may bring relevant information that can be used to clarify the evolutionary history of selenoproteins among vertebrates and permit to accurately set the filogenetical position of Crysemys picta belli.

Resum

El genoma de la Chrysemys picta belli ha estat seqüenciat recentment. L'objectiu principal d'aquest treball és trobar en el genoma tots els homòlegs de selenoproteïnes coneguts. Les selenoproteïnes són una família de proteïnes que es caracteritza per contenir la selenocisteïna, el 21è aminoàcid. Aquestes proteïnes tenen una funció essencial en nombrosos processos cel·lulars com la protecció davant l'estrés oxidatiu o l'activació de la resposta immunitària. La selenocisteïna és codificada per un codó TGA, aquest codó en condicions normals és un codó de STOP. Per a la traducció de les selenoproteïnes són necessàris certs elements com els SECIs, així com diversos enzims. La particularitat d'aquest model de traducció és un greu problema que dificulta la correcta anotació de les selenoproteïnes. Hem dissenyat un conjunt seriat de programes que integren diverses eines com: BLAST, Exonerate, GeneWise, T-Coffee, SECISearch i BLAST contra non-redundant NCBI database amb l'objectiu d'identificar i anotar totes les proteïnes existents en el genoma de Crysemys picta belli. Els resultats poden aportar informació rellevant que pugui ser útil per desxifrar la història evolutiva de les selenoproteïnes en els vertebrats, al mateix temps que permeti acurar la posició filogenètica de la Crysemys picta belli.
